G-418 is a member of the large family of Aminoglycoside antibiotics and has a molecular structure almost like Gentamycin. The antibiotic is an irreversible inhibitor of protein synthesis. Due to its structure is capable of binding to both prokaryotic and eukaryotic ribosomes and inhibiting protein synthesis and generating errors in the transcription of the genetic code. G-418 can by inactivated by NPTII or 3APH. The hydroxyl group present at the 3 position of G-418 is phosphorylated by NPTII. The resulting steriometric change of the molecular structure of the antibiotic prevents it from binding at its ribosome binding site. -

MES, 2-(N-morpholino)ethanesulfonic acid contains a morpholine ring. Its molecular weight is 195.2 and the chemical formula is C6H13NO4S. MES is used as a buffering agent in biology and biochemistry. It has pKa value of 6.15 at 20 °C. The pH (and pKa at ionic strength I≠0) of the buffer solution changes with concentration and temperature. MES is highly soluble in water. MES was developed as one of Good's buffers in the 1960s. These buffers were developed with the following criteria in mind: midrange pKa, maximum water solubility and minimum solubility in all other solvents, minimal salt effects, minimal change in pKa with temperature, chemically and enzymatically stable, minimal absorption in visible or UV spectral range and reasonably easily synthesized. -

MOPS (3-(N-morpholino)propanesulfonic acid, 4-Morpholino Propanesulfonic Acid) is a buffer introduced by Good et al. in the 1960s. It is a structural analog to MES and it’s chemical structure contains a morpholine ring. With a pKa of 7.20, MOPS is an excellent buffer for many biological systems at near-neutral pH. MOPS has inert properties and is capable of buffering within a pH range of 6.5 – 7.9 in solutions. MOPS does not interact with any metal ions, and with it’s significant metal-buffer stability it is especially non-reactive with cobalt (Co), copper (Cu), manganese (Mn), nickel (Ni) and zinc (Zn). MOPS is frequently used as a buffering agent in biology and biochemistry. It has been tested and recommended for polyacrylamide gel electrophoresis. Usage above a 20 mM concentration in mammalian cell culture work is not recommended.
