The uptake of Canavanine leads to incorporation in protein in the place of Arginine, thereby producing structurally aberrant proteins that may not function properly. Both Canavanine and Arginine are imported into the cell by the same high affinity permease which is encoded by the CAN1 locus. G-418 can by inactivated by NPTII or 3�APH. The hydroxyl group present at the 3� position of G-418 is phosphorylated by NPTII. The resulting steriometric change of the molecular structure of the antibiotic prevents it from binding at its ribosome binding site.
